LEUKEMIA PANEL IMMUNOPHENOTYPING BY FLOW CYTOMETRY

Guidelines

This panel is designed to distinguish between acute lymphoid and non-lymphoid leukemia.

Specimen Collection Requirements

Collect

Bone Marrow Aspirate, collected in a heparinized syringe and transfer into an EDTA tube or into RPMI for transport to the lab, also include 2 unstained aspirate smears. If a large percentage of blasts are present in the peripheral blood, Whole Blood in a lavender (EDTA) top tube is preferred, but heparin may also be used.

Handling

Maintain specimens at room temperature, avoid temperature extremes and do not centrifuge. Call Client Services ASAP at (503) 413-1234 or Toll Free (877) 270-5566 to arrange for PRIORITY pickup.

Always submit recent CBC (collected within past 72 hours), differential and 2 unstained blood smears with request, for both bone marrow or blood specimens.

Preferred Volume

Bone Marrow: 4.0 mL, Blood: 4.0 mL

Minimum Volume

Bone Marrow: 0.5 mL, Blood: 2.0 mL, must have sufficient number of malignant cells for analysis

Transport

Room temperature (20-25°C)

Rejection Criteria

BONE MARROWS: Never rejected without consultation with Hematopathologist. BLOOD: Clotted, severely hemolyzed or frozen specimens

Stability

Ambient: 24 hours; Refrigerated: 24 hours; Frozen: Unacceptable
