Guidelines

Array-based comparative genomic hybridization is becoming a widely used approach for the detection of copy number variations, especially in children referred for unexplained mental retardation and/or multiple congenital anomalies.

Specimen Collection Requirements

Collect

Whole Blood Adult: Dark Green (Sodium Heparin) and Lavender (EDTA) 3-5 mL each tube.

Whole Blood Infants (Newborns ≤ 1 month of age): Dark Green (Sodium Heparin) AND Lavender (EDTA) 0.5-1.0 mL each tube. Microtainer, capillary, or finger/ heel stick collections are NOT accepted.

Cord Blood: Dark Green (Sodium Heparin) AND Lavender (EDTA) 0.5-1.0 mL each tube.

Amniotic Fluid: Minimum of 15 mL in a sterile tube.

Chorionic Villi: 10-15 mg sample obtained and transported aseptically in sterile medium; MEM, RPMI or saline. The transport container must be sterile.

Solid Tissue: 20 mg (4 mm x 4 mm), sample obtained and transported aseptically in sterile medium; MEM, RPMI or saline. The transport container must be sterile. Do NOT place in formalin.

Solid Tumor: 20 mg (4 mm x 4 mm), sample obtained and transported aseptically in sterile medium; RPMI or saline. The transport container must be sterile. Do NOT place in formalin.

Handling

Do not refrigerate, freeze or expose to extreme temperatures. Keep specimen at ambient temperature and transport within 24 hours. Maintain sterility, do not split specimen or open tube.

DO NOT PLACE IN FORMALIN.

Preferred Volume

Whole Blood              Infants: 1.0 mL each tube (no heel sticks)

                                      Others: 3.0-5.0 mL in each tube

Cord Blood                 1.0 mL each tube

Amniotic Fluid            20 mL in sterile tube

Chorionic Villi            15 mg sample

Solid Tissue               20 mg sample

Minimum Volume

Whole Blood              Infants – 0.5 mL in each tube

                                      Others – 1.0 mL in each tube

Cord Blood                 0.5 mL each tube

Amniotic Fluid            15 mL in sterile tube

Chorionic Villi             7 mg sample

Solid Tissue               20 mg sample

Transport

At room temperature within 24 hours

Rejection Criteria

Capillary sticks, finger/heel sticks and specimens in formalin.

Specimens should only be rejected by Cytogenetics staff.
