Guidelines

Qualitative measurement of Herpes simplex antibodies. Almost all recurrent genital herpes cases are caused by HSV type 2, yet HSV type 1, the causative agent of orolabial herpes, causes up to 30% of first episode genital herpes.

Specimen Collection Requirements

Collect

Serum, one 5.0 mL gold (SST) or 7.0 mL red top tube

Handling

Allow serum to clot completely at room temperature (minimum – SST: 30 minutes, red top tubes: 60 minutes). Centrifuge and separate serum from cells ASAP.

Preferred Volume

0.5 mL Serum

Minimum Volume

0.25 mL Serum (0.75 mL minimum whole blood draw)

Refrigerated (2-8°C)

Rejection Criteria

Grossly hemolyzed, icteric or lipemic specimens; contaminated specimens; heat inactivated specimens; plasma.

Stability

AFTER SEPARATION FROM CELLS:  Room temperature (18-26°C): Unacceptable; Refrigerated (2-8°C): 7 days; Frozen (≤ -10°C): 30 days (not more than 5 freeze/thaw cycles).

>= 1.10 Index          Positive for IgG antibody to HSV type 1 glycoprotein G.

HSV-2 IgG
<= 0.89 Index          Negative for IgG antibody to HSV type 2 glycoprotein G.  Repeat testing in 4-6 weeks may be helpful.
