Guidelines

The detection of IgG antibody to measles can be used to indicate Immune Status.

Specimen Collection Requirements

Collect

Serum, one 5.0 mL gold (SST) or 7.0 mL red top tube

Handling

Allow serum to clot completely at room temperature (minimum – SST: 30 minutes, red top tubes: 60 minutes). Centrifuge and separate serum from cells ASAP.

Preferred Volume

0.5 mL Serum

Minimum Volume

0.25 mL Serum (0.75 mL minimum whole blood draw)

Transport

Refrigerated (2-8°C)

Rejection Criteria

Grossly hemolyzed, icteric or lipemic specimens; contaminated specimens; heat inactivated specimens; plasma.

Stability

AFTER SEPARATION FROM CELLS: Room temperature (18-26°C): Unacceptable; Refrigerated (2-8°C): 9 days; Frozen (≤ -10°C): 23 days (not more than 6 freeze/thaw cycles).

Reference Values

< 13.5 AU/mL            Negative for measles (rubeola) IgG antibody.

 

13.5 – 16.4 AU/mL    Equivocal for measles (rubeola) IgG

                                  antibody. Repeat testing in 7 – 14 days

                                  may be helpful.

 

≥ 16.5 AU/mL            Positive for measles (rubeola) IgG antibody. 

                                  A positive result generally indicates exposure to

                                  measles virus or previous vaccination.

 

NOTE: The best evidence for current infection is a significant change on two appropriately timed specimens, where both tests are done in the same laboratory at the same time.same laboratory at the same time.
